Materials Required:
- HyperSilMag Viral RNA Kit
- Viral sample (serum, plasma, swab fluid)
- Carrier RNA (provided)
- Lysis/Binding Buffer (provided)
- Wash Buffers 1 & 2 (provided)
- Elution Buffer (provided)
- Magnetic separator
Procedure:
- Sample Preparation: Add 140 uL viral sample to 560 uL Lysis/Binding Buffer containing carrier RNA. Mix thoroughly.
- Binding: Add 20 uL magnetic beads. Incubate 10 minutes at room temperature with gentle mixing.
- Magnetic Separation: Place on magnetic separator for 1-2 minutes. Carefully remove and discard supernatant.
- Wash 1: Add 500 uL Wash Buffer 1. Mix gently, separate, and discard supernatant.
- Wash 2: Add 500 uL Wash Buffer 2. Repeat wash step twice.
- Drying: Air-dry beads on magnetic separator for 3 minutes.
- Elution: Add 60 uL Elution Buffer. Incubate at room temperature for 2 minutes. Magnetically separate and transfer RNA to clean tube.
Yield: > 95% recovery for viral loads >=100 copies/mL
Time: Total protocol time approximately 30 minutes
