Materials Required:
- HyperSilMag-Epoxy beads
- Target molecule with amino, thiol, or hydroxyl groups
- Coupling buffer (0.1 M sodium phosphate pH 8.5-9.5)
- Wash buffer (PBS)
- Blocking solution (1 M Tris-HCl pH 8.0)
Procedure:
- Bead Washing: Wash epoxy beads 3횞 with coupling buffer.
- Coupling: Add target molecule (1-5 mg/mL) in coupling buffer. Incubate overnight at room temperature or 4-8 hours at 37째C with gentle rotation.
- Monitoring: Epoxy groups react with -NH2 (fastest), -SH (moderate), and -OH (slower). Adjust pH 8.5-9.5 for optimal amine coupling.
- Blocking: Add Tris blocking solution. Incubate 2-4 hours at room temperature to block remaining epoxy groups.
- Washing: Wash thoroughly 4횞 with PBS to remove unbound material and excess blocking agent.
- Storage: Store at 4째C in PBS with 0.02% sodium azide.
