Materials Required:
- HyperSilMag-Carboxyl beads
- Target protein or amine-containing molecule
- EDC (50 mg/mL in water, prepare fresh)
- NHS (50 mg/mL in water)
- MES buffer pH 5.5
- PBS pH 7.4
Procedure:
- Bead Preparation: Wash carboxyl beads 3횞 with MES buffer pH 5.5.
- Activation: Add EDC (10 mg/mL final) and NHS (10 mg/mL final). Mix 15 minutes at room temperature.
- Washing: Wash activated beads 2횞 with cold MES buffer to remove excess EDC/NHS.
- Coupling: Add target molecule (0.5-2 mg/mL) in PBS pH 7.4. Incubate 2-4 hours at room temperature with rotation.
- Blocking: Add ethanolamine pH 8.5 to 50 mM final concentration. Incubate 30 minutes.
- Final Wash: Wash 3횞 with PBS. Store conjugated beads at 4째C.
Coupling Yield: Typically >80% for most proteins
