Carboxyl Bead Conjugation Protocol

Materials Required:

  • HyperSilMag-Carboxyl beads
  • Target protein or amine-containing molecule
  • EDC (50 mg/mL in water, prepare fresh)
  • NHS (50 mg/mL in water)
  • MES buffer pH 5.5
  • PBS pH 7.4

Procedure:

  1. Bead Preparation: Wash carboxyl beads 3횞 with MES buffer pH 5.5.
  2. Activation: Add EDC (10 mg/mL final) and NHS (10 mg/mL final). Mix 15 minutes at room temperature.
  3. Washing: Wash activated beads 2횞 with cold MES buffer to remove excess EDC/NHS.
  4. Coupling: Add target molecule (0.5-2 mg/mL) in PBS pH 7.4. Incubate 2-4 hours at room temperature with rotation.
  5. Blocking: Add ethanolamine pH 8.5 to 50 mM final concentration. Incubate 30 minutes.
  6. Final Wash: Wash 3횞 with PBS. Store conjugated beads at 4째C.

Coupling Yield: Typically >80% for most proteins
