Biotin-Tagged Molecule Capture Protocol

Materials Required:

  • HyperSilMag Biotin
  • Streptavidin-conjugated target molecule (protein, nucleic acid, etc.)
  • Binding Buffer (PBS with 0.05% Tween-20)
  • Wash Buffer (PBS with 0.1% Tween-20)
  • Elution Buffer (optional, for recovery)
  • Magnetic separator

Procedure:

  1. Bead Preparation: Wash Biotin Beads 2x with Binding Buffer using magnetic separation.
  2. Binding: Resuspend beads in Binding Buffer. Add streptavidin-conjugated target (0.1-10 ug/mL). Incubate 15-30 minutes at room temperature with gentle rotation.
  3. Washing: Magnetically separate and wash 3x with Wash Buffer to remove unbound material.
  4. Application: Use beads directly for assays, or proceed to elution if target recovery is needed.
  5. Elution (Optional): Add Elution Buffer (e.g., biotin solution 5 mM). Incubate 10 minutes with mixing. Magnetically separate and collect eluate.

Binding Capacity: >3000 pmol biotin per mg beads

Affinity: Kd = 10^-15 M (one of the strongest non-covalent interactions)
