Amino Bead Conjugation Protocol

Materials Required:

  • HyperSilMag-Amino beads
  • Target molecule with carboxyl or carbonyl groups
  • Coupling reagent (e.g., glutaraldehyde for proteins)
  • Activation buffer (pH 7-8)
  • Wash buffer (PBS)
  • Blocking solution (Tris or ethanolamine)

Procedure:

  1. Bead Washing: Wash amino beads 3x with activation buffer.
  2. Activation: For protein coupling, add 2.5% glutaraldehyde solution. Incubate 1-2 hours at room temperature.
  3. Washing: Remove glutaraldehyde and wash thoroughly with PBS.
  4. Coupling: Add target molecule (0.1-1 mg/mL). Incubate overnight at 4°C or 2-4 hours at room temperature with rotation.
  5. Blocking: Add blocking solution (1 M Tris, pH 8.0). Incubate 30-60 minutes to block unreacted amino groups.
  6. Final Wash: Wash 3x with PBS. Store at 4°C in PBS with 0.02% sodium azide.

Coupling Efficiency: Typically 70-90% for most biomolecules
